Research Update

The Challenge of Targeted Delivery

A new challenge - the synthesis of folate-AMP, a key compound in a new pRNA/siRNA drug delivery system - was recently presented to TriLink because of our expertise in unique nucleic acids. So, what is pRNA? pRNA is employed by bacteriophages during replication to pack their DNA into protein shells. Zhang et.al. used pRNA to specifically target cells without the need for transfection. In this study pRNA was covalently bound to an siRNA drug and fluorescently-tagged folate to target HeLa cell folate receptors. The unique compound, folate-AMP, needed to label the pRNA was manufactured by TriLink.

TriLink offers an array of custom chemistry services for your unique research needs including custom nucleoside mono-, di- and triphosphates, phosphoramidites and other small molecules. Contact us today to discuss your project.

Technology Spotlight

Your only source for ppGpp

Guanosine-3', 5'-Bisdiphosphate (ppGpp) is involved in RNA synthesis inhibition, an adaptive response of bacteria to amino acid starvation. In fact, there are many genes up-regulated by ppGpp during a cell’s stress response. ppGpp is commonly used to research the inhibition, rates and overall control of transcription and elongation of bacterial RNA.

TriLink is the only source for many unique modified nucleotides, including ppGpp. Leading the industry in modified nucleic acids means continually advancing our science. Recently we made a significant breakthrough in the production of ppGpp, greatly improving the yield and purity of the intermediate. We are now able to offer an uninterrupted supply.

Question of the Month

Q - Can TriLink prepare custom phosphoramidites and polyphosphates under GMP conditions?

A - We can prepare any kind of nucleotide in our GMP facility. We first develop, if necessary, the process in our R&D lab, then validate it in our GMP facility with our equipment and reagents. All TriLink products are GMP and manufactured in compliance with ISO 9001:2008.
