Oligonucleotide Purification

TriLink offers a selection of purification processes for you to choose from. Note some modified oligonucleotides can only be isolated using particular processes. We will inform you if the method you have chosen will not give you the quality you seek or if we have other suggestions.

These prices also apply to base modified and conjugated oligonucleotides, although additional fees may be required in cases where an additional purification is a necessary.

In general, yields of HPLC purified (~ 90%) unmodified DNA oligonucleotides are as follows:

0.2 umole scale: 5 - 15 OD units (~0.15 mg - 0.5 mg)
1.0 umole scale: 20- 60 OD units (~0.66 mg - 2 mg)
15 umole scale: 300 - 750 OD units (~10 - 25 mg)

Yield and purity differences can be caused by many factors, such as sequence, length and composition.
 
RNA (2' OH) Oligonucleotides

In general, yields of PAGE purified (>90%) unmodified RNA oligonucleotides are as follows:

0.2 umole scale: 2 - 8 OD units (~0.07 mg - 0.27 mg)
1.0 umole scale: 10 - 40 OD units (~0.33 mg - 1.33 mg)
15 umole scale: 150 - 400 OD units (~4.45 mg - 13.33 mg)

2' Modified Oligonucleotides (2' Fluoro and 2' Amino)
These modified oligonucleotides can often be purified using the same methods shown for DNA, however, much depends on the rest of the molecule. Likewise, yields of these molecules are extremely variable, dependent on the design of the entire molecule, and are very sequence specific. Please contact us for more information regarding purification or yields.
