Alpha thiol nucleotides were first introduced over 30 years ago and remain a valuable molecular biology tool. The foremost applications of these nucleotides are imparting nuclease resistance to an aptamer and introducing nicking sites in nick translation.
Nick translation is one of the most common methods for introducing labeled nucleotides into a double stranded DNA molecule. It involves making small nicks in one strand of the DNA with DNAse and subsequently incorporating thio modified nucleotides by repairing them with DNA polymerase I. When alpha thio modified dNTPs are used in PCR, phosphorothioate-based sequencing is possible.
