CleanAmp™ Master Mixes

CleanAmp™ PCR Master Mixes are powered by TriLink's novel, heat-activated dNTP chemistry. Activation of the CleanAmp™ dNTPs occurs during the initial denaturation of Hot Start PCR and each subsequent thermal cycle, releasing just enough reagent to allow efficient amplification. By limiting the amount of active reagent during the early cycles when the target is in low concentration, CleanAmp™ significantly reduces background amplification to achieve a reduction or even an elimination of primer dimer and mis-priming.

CleanAmp™ One-Step RT-PCR 2X Master Mix

The CleanAmp™ One-Step RT-PCR 2X Master Mix is an innovative solution employing TriLink’s patented CleanAmp™ technology. CleanAmp™ enables a Hot Start at both the RT and PCR steps, significantly improving the specificity of one-step reverse transcription PCR. Perform end-point or real-time RT-PCR assays with RNA input levels as low as 32 picograms.

As shown in the figure below, Hot Start control of the PCR step alone cannot effectively suppress off-target amplifications. Introduction of the CleanAmp™ technology provides an elegant solution by avoiding the complexity and cost of integrating a Hot Start solution for both the RT and the PCR step. The CleanAmp™ One-Step RT-PCR 2X Master Mix is compatible with end-point PCR, real-time and multiplex PCR.

CleanAmp™ Multiplex PCR 2X Master Mix

CleanAmp™ Multiplex PCR 2X Master Mix is an optimized, ready-to-use mix of CleanAmp™ dNTPs and Taq DNA Polymerase in reaction buffer suited for multiplex amplification. CleanAmp™ Mulitplex PCR 2X Master mix has been utilized in assays of 50 targets and higher. Simply add primers, template DNA and water.

CleanAmp™ GC-Rich PCR 2X Master Mix

The CleanAmp™ GC-Rich PCR 2X Master Mix is optimized specifically for the amplification of difficult GC-rich targets with up to 85% GC content. It is a convenient, ready-to-use 2X Hot Start reaction pre-mix which combines TriLink’s CleanAmp™ 7-deaza-dGTP Mix and Taq DNA polymerase. Simply add primers and template DNA.

CleanAmp GC-Rich Target Amplification

CleanAmp™ GC-Rich PCR 2X Master Mix provides robust amplification for targets with 60% GC content or greater. Three targets ranging from 80 to 85% GC content were amplified from 5 ng of Human Genomic DNA.

CleanAmp™ GC-Rich PCR 2X Master Mix improves PCR sensitivity and efficiency over a range of input target concentrations. (A) Agarose gel analysis of the experiment. (B) Standard curve from the real-time PCR experiment, with hydrolysis probe-based detection. The GNAQ target with a GC content of 79% was amplified from 0.8 to 500 ng input Human genomic DNA in a real-time PCR protocol where the CleanAmp™ GC-Rich PCR 2X Master Mix was compared to analogous mixes prepared with a standard dNTP mix or a standard 7-deaza-dGTP mix.

 

CleanAmp™ GC-Rich PCR 2X Master Mix can be adapted for use in real-time PCR protocols by the addition of the appropriate dyes and/or probes for detection. Cloning and Sanger dideoxy sequencing are two downstream applications that benefit from the use of this master mix. GC-rich targets PCR amplified with the CleanAmp™ GC-Rich PCR 2X Master Mix prior to Sanger dideoxy sequencing experience reduced background noise and higher Phred quality scores throughout the entire sequencing trace.

 

CleanAmp™ GC-Rich PCR 2X Master Mix improves downstream Sanger dideoxy sequencing results over the entire length of the B4GN4 (Beta-1,4-N-acetyl galactosaminyltransferase) target. The 78% GC-rich target was PCR amplified from 5 ng of Human genomic DNA using either the CleanAmp™ GC-Rich 2X PCR Master Mix or an analogous mix prepared with a standard 7-deaza-dGTP mix. The resulting amplicons were submitted for Sanger dideoxy sequencing.
