Developing Antibodies Using Oligonucleotide-directed Mutagenesis

Commonly used to improve the specific biological function of proteins, oligonucleotide-directed mutagenesis has been particularly relevant in monoclonal antibody development, as demonstrated in a recent article by H. Karauzum and colleagues, where the authors not only utilized this technology to develop highly specific and effective antibodies towards Staphylococcal Enterotoxin B (SEB) but, quite interestingly, demonstrated that these antibodies can be produced in a plant expression system.

Trimers for Codon Mutagenesis

Trimer incorporated oligos have become increasingly popular, especially in antibody development, for achieving base variability with specific ratios of amino acid mutations. The use of trimers allows the randomization of specific sites to yield specific amino acids and to avoid stop codons and sequence repeats. TriLink is the leading provider of trimer-incorporated oligos.
