Genome-scale CRISPR Screens Identify Critical Host Factors for Hepatitis C, Dengue, Zika and Other Mosquito-borne Flaviviruses

In a recent Nature paper, researchers at Stanford University led by Caleb Marceau reported using advanced high-throughput screening methods to identify novel host factors that are important for viral infection and replication of Flaviviridae viruses. Flaviviridaes are a family of single stranded RNA viruses that cause illness in millions of people worldwide each year and includes the Dengue virus, Hepatitis C virus, and Zika virus (for more on the Zika virus, see this month's Zone in with Zon blog.)

Using reporter mRNA from TriLink, the investigators are identifying new avenues for targeted drug development against these viruses in order to better understand the host factors that help these viruses infect and replicate.

TriLink's Reporter Gene mRNA

Reporter mRNAs can be used as controls to study mRNA transfection and expression in mammalian cells using a variety of assays including fluorescence microscopy, quantitative fluorometry and bioluminescent imaging, as well as fluorescence-activated cell sorting (FACS).

TriLink offers reporter gene mRNA with a variety of the most popular standard and custom base modifications patterns to:

  • Reduce innate immune response
  • Enhance Expression
  • Directly visualize mRNA delivery

How much mRNA is needed to transfect cell cultures in 24-well plates?

There are typically two avenues for transfection of mRNA depending on cell type:
1. For adherent cells, use a transfection reagent such as MessengerMax (Invitrogen), mRNA TransIT (Mirus) or mRNA-In (MTI-GlobalStem).....
