BspQI (isoschizomers: SapI, PciSI and LguI) is a Type IIS restriction enzyme known for its unique cleavage properties. It recognizes asymmetric DNA sequences, cleaves the double-stranded DNA outside of the recognition site, and generates a 5′ overhang.
BspQI’s recognition and cleavage sites:
5' GCTCTTCN↓ 3'
3' CGAGAAGNNNN↑ 5'
BspQI is thermostable and can be used in various molecular biology applications, including generation of linear DNA templates for in vitro transcription to synthesize RNA.
The enzyme is provided 10 U/μL in storage buffer. It is also supplied with 1.25 mL of 10X RE Reaction Buffer (500 mM Tris,1000 mM NaCl, 100 mM MgCl2, 1 mg/mL rHSA, pH 7.5 at 25°C).
Unit definition: The amount of BspQI required to digest 1 µg of lambda DNA in 1 hour at 50°C in a 50 µL reaction.
